Positive and Negative Regulation of Muscle Cell Identity by Members of the Hedgehog and TGF-Î2 Gene Families

چکیده
We have examined whether the development of embryonic muscle fiber type is regulated by competing influences between Hedgehog and TGFb signals, as previously shown for development of neuronal cell identity in the neural tube. We found that ectopic expression of Hedgehogs or inhibition of protein kinase A in zebrafish embryos induces slow muscle precursors throughout the somite but muscle pioneer cells only in the middle of the somite. Ectopic expression in the notochord of Dorsalin-1, a member of the TGFb superfamily, inhibits the formation of muscle pioneer cells, demonstrating that TGFb signals can antagonize the induction of muscle pioneer cells by Hedgehog. We propose that a Hedgehog signal first induces the formation of slow muscle precursor cells, and subsequent Hedgehog and TGFb signals exert competing positive and negative influences on the development of muscle

متن کاملGene Regulation Network Based Analysis Associated with TGF-beta Stimulation in Lung Adenocarcinoma Cells
Background: Transforming growth factor (TGF)-β is over-expressed in a wide variety of cancers such as lung adenocarcinoma. TGF-β plays a major role in cancer progression through regulating cancer cell proliferation and remodeling of the tumor micro-environment. However, it is still a great challenge to explain the phenotypic effects caused by TGF-β stimulation and the effect of TGF-β stimulatio...

Vertebrate skeletal muscles contain muscle fibers of several types, which can be broadly classified as slow or fast fibers, on the basis of differences in contraction speeds, metabolic activities, and motoneuron innervation. We have examined the potential roles of growth factors of members of the hedgehog and TGF-ß gene families in the formation of slow muscle fibers including muscle pioneer (M...

Background: Despite substantial progress in the clinical management of diabetes, diabetic nephropathy (DN) still occurs recurrently, implicating diabetes as the major underlying condition leading to the end stage renal disease. One of the main reasons is the influential role of genetic or inherited backgrounds of diabetics that are almost overlooked in daily practice. Owing to be orchestrated b...
متن کاملGene Family: Structure, Organization and Evolution
Gene families are considered as groups of homologous genes which they share very similar sequences and they may have identical functions. Members of gene families may be found in tandem repeats or interspersed through the genome. These sequences are copies of the ancestral genes which have underwent changes. The multiple copies of each gene in a family were constructed based on gene duplicati...
